On day 2, postconfluent cells were incubated in DMEM + 10% FBS (control) in the presence of ML-7 (6 × 10−5 M; ML-7) and in the presence of both calyculin A (2 × 10−7 M) and ML-7 (6 × 10−5 M; ML-7 + Caly) for 1 h. The level of pMLC20 was determined using supernatant protein (80 μg).
